Chemically Rizatriptan Benzoate is the N, N-diethyl -5(1H-1,2,4-triazol-1-ylmethyl)-1H-indole-3-ethanamine mono benzoate and is a selective 5-hydroxytriptamin 5-HT] 1B/1D receptor agonist. It has a weak affinity for other 5-HT receptor subtype and was launched in 1998 for the acute treatment of migraine in adults. It has good water solubility (42mg/ml) and poor bioavailability 45%] due to hepatic first pass effect. The suggested mechanism of anti-migraine action of Rizatriptan Benzoate is through activation of post synaptic 5-HT 1B receptor within cerebral and Dural vessel walls, causing vasoconstriction of per vascular nerve terminals. The most prominent advantage of in situ gel over the conventional gel is that it's can be easily instilled as a drop allowing accurate drug dosing. The objectives of presented study is to develop in situ gel of Rizatriptan Benzoate, with favourable gelation, viscosity, temperature and released property. This may gives patient friendly, needle free dosage form.
