Preservation of DNA at Room Temperature After Exctraction

DNA extracted from Escherichia coli samples is the essence of genomic and translational research. However, few options other than cryogenic storage are available in C te d'Ivoire for the preservation of genetic material. Cryogenic storage is expensive in terms of energy, equipment, and maintenance, and genetic material is generally subject to contamination and degradation in the event of a power failure or outage. The overall objective of this study was to evaluate the stability of genetic material and the persistence of resistance genes detected by antibiogram after storage at room temperature. To do this, 38 strains of Escherichia coli isolated from cattle and poultry slaughterhouse effluents were used. The level of antibiotic resistance of the strains was determined using an antibiogram. Two types of extraction were performed, one with guanidine thiocyanate and the other with the Qiagen Kit, followed by DNA quantification before and after storage at different temperatures, namely room temperature and liquid nitrogen.
